A Hyundai SUV collided with a Mitsubishi sedan at Hackham on Main South Road in the early hours of Saturday morning.
Georgina Heath was found dead at the scene in the SUV, while two other passengers — a 28-year-old woman and a 30-year-old man — were taken to Flinders Medical Centre in serious but not life-threatening conditions.
The 35-year-old driver of the SUV was in a similarly serious condition and the 17-year-old driver of the Mitsubishi and his passenger sustained serious injuries in the crash.
South Australian Police are investigating the incident.
The woman's death marks the 34th life lost on South Australian roads this year.
