Police said the 14-year-old boy was walking home from a night out with two friends when a vehicle came out of nowhere and struck the victim.
Two offenders armed with knives got out of the vehicle, before assaulting him and leaving him on the road in St Albans about 11pm, police say.
Emergency services found the Braybrook boy lying on the road. He died while being transported to hospital.
His friends fled the scene and contacted police immediately.
Detective Inspector Dave Dunstan said the attack was "targeted".
"We have no information at this stage that it's gang related, but we believe this was a targeted assault on the victim," he told media today.
"We don't know the relationship at this stage, but the victim and his friends were walking on the street when this vehicle came as a surprise from nowhere."
It comes as CCTV has emerged of a group of three teenagers, believed to be the victim and his friends before the attack.
The footage was captured from a row of shops close to Ginifer train station.
Neighbours living in the suburb, about 17km from Melbourne's CBD, said they were woken up after hearing commotion outside.
"I heard a car door shut and then I heard screaming and people shouting," one resident said.
"(It's) a bit dangerous to walk here in the night."
A crime scene has been established while officers work to determine the cause of the teen's death.
Homicide Squad detectives are on the scene as they investigate the boy's death.
Officers from the Major Crime Scene Unit are at the scene.
