The 14-year-old girl allegedly driving at the time is under police guard while the other passenger, a 12-year-old boy is recovering from minor injuries.
Superintendent Michael Layton alleged the teens were seen to be "driving erratically" before the collision along the Hume Highway in Locksley, with the truck driver exclusively speaking to 9News about his ordeal.
The car collided with the truck near Alexandersons Road about 8.30am, killing the 15-year-old front-seat passenger.
Police allege the wagon was stolen from a driveway near Wangaratta last night.
"This is a tragic scene and high-risk taking behaviour, it's totally avoidable.. and is every parent's worst nightmare," Superintendent Layton said.
"It's a life-changing event for the family of the deceased 15-year-old girl and also the the family of other two youths."
The 45-year-old B-double truck driver from Tabilk was not injured and is assisting police.
Clint Seddon told 9News the incident "hit him hard".
"One is dead and two have to suffer for the rest of their lives," he said.
The 14-year-old driver remains in hospital with non-life-threatening injuries and is under police guard.
Locksley is about 139km north of Melbourne and is near the regional hub Shepparton.
Police are investigating and remain on the scene.
