The pair died after two off-road motorbikes collided on Beattys Road in Grangefields near Melton about 6.35pm yesterday.
The family was celebrating a children's birthday party held on Father's Day with loved ones when the crash occurred.
Detective Senior Sergeant Paul Lineham said police believed the teens were riding two unregistered motorbikes when they collided head-on.
Zena, 18, died instantly while Ali, 17, was taken to hospital but later died.
Two boys, aged six and 11, were also rushed to the Royal Children's Hospital with serious injuries.
The children remain in hospital in a stable condition.
Lineham said it was unknown if the injured children were riding on the back of the bikes or if they were bystanders.
"Unfortunately we haven't received much with cooperation with the family and they were taken by relatives to hospital," he said.
"It was a public road, neither were licensed for these motorbikes and the bikes were unregistered."
Zena's father, Ali Houli, was rushed to hospital after he jumped on a quad bike to help his daughter after the crash.
The 43-year-old Altona Meadows man lost control, collided with a speed sign and crashed near the scene of the other accident.
Omar El Houli, the uncle of the two teenagers, and owner of the property said the loss was "heartbreaking".
"We were driving down (the road) and we got out. My niece was on the floor dead and my nephew was also on the ground," El Houli told media today.
"I wanted to see who they were, it was my farm. I saw the motorbikes, I saw them, they were on the ground.
"I couldn't imagine anything worse than what happened."
El Houli said he was still trying to piece together how the incident occurred.
"My niece was an angel and so was my nephew, they were the best," he said.
"There was a few of us here with families getting together, I don't exactly know what happened.
"I wish I could put my life there instead, they were beautiful kids.
"I can't explain it, family is all you got, if you can't look after them - what do you got?"
Neighbour Tom Panza said there was poor visibility at the time of the crash.
"They were having a bit of fun and party and enjoying themselves during the day, but (as) the sun was going down and I saw a few bikes going up and down," he told media.
"There were no lights and I thought something might happen.
"Unfortunately, it did."
Police are investigating the circumstances around the crash.
